How are you expecting to acquire the sound? Using the audiorecorder, DSP audiorecorder, or the Data Acquisition Toolbox?

You can either use the Data Acquisition Toolbox or the audiorecorder function. Check the examples in script3.m and script4.m of my audio analysis library, part of the book Introduction to audio analysis .
Both examples demonstrate how to use acquisition + processing of the recorded data.
