Does Matlab need App Designer or any toolbox to run an app created using App Designer?

App Designer apps do not depend on App Designer to run, but the apps use features that were first introduced in R2016a, which is the same release App Designer was released. So if a MATLAB doesn't have App Designer, it means it's a pre-R2016a release, in which case it definitely won't have features App Designer apps require.

Yes, it is part of MATLAB from R2016a onwards.
